ABSTRACT: Sauger are an endemic fish species located in two distinct geographical areas in Wisconsin, the Mississippi River drainage and the Lake Michigan drainage. They are found in the tributaries of the Mississippi River drainage, the St. Croix, Black, Chippewa, Wisconsin Rivers and the tributaries of the Lake Michigan drainage, including Green Bay, Lake Winnebago, and the Fox, and Wolf Rivers. The objective of this study was to determine if genetic differences are present in native populations of Sauger observed in Lake Winnebago within the Lake Michigan drainage and the Lower Wisconsin River within the Mississippi River drainage. Fin clips from 199 adult Sauger from the Lower Wisconsin River and 82 adult Sauger from Lake Winnebago were collected for genetic analysis. These samples were genotyped with a standard suite of ten microsatellite loci. Genetic data from the two Sauger populations were analyzed using the programs GenAlEx and STRUCTURE.  Genetic differentiation calculated using pairwise FST values was 0.045 between populations, indicating higher differences than the Walleye populations from the same locations (FST=0.018). STRUCTURE results confirmed the observation of two distinct genetic groups.  Our results demonstrate that significant genetic structure exists between Sauger populations in the Mississippi River and Lake Michigan drainages.  We suggest using local broodstock in future stocking efforts to ensure that these important genetic differences are maintained.Citation: B. If future stocking efforts of Sauger are to take place in Wisconsin to supplement the recreational fishery, genetic analysis should be considered to prevent the mixture of distinct genetic stocks.
